Gr\u00e2ce \u00e0 une \u00e9tude d\u2019association pang\u00e9nomique (GWAS), nous avons r\u00e9cemment identifi\u00e9 un locus dans le g\u00e9nome de <em>M. larici-populina<\/em>, qui pourrait correspondre au locus d\u2019avirulence Avr7, dont la mutation est responsable du contournement de la r\u00e9sistance R7 du peuplier. L\u2019objectif de la th\u00e8se sera d\u2019\u00e9tudier cette r\u00e9gion g\u00e9nomique et de valider fonctionnellement le locus d\u2019int\u00e9r\u00eat. Il s\u2019agirait alors du premier g\u00e8ne d\u2019avirulence identifi\u00e9 chez ce champignon. Plusieurs approches compl\u00e9mentaires sont envisag\u00e9es\u00a0: (<em>i<\/em>) une approche de g\u00e9nomique comparative afin de pr\u00e9ciser le polymorphisme dans cette r\u00e9gion\u00a0; (<em>ii<\/em>) une approche de transcriptomique afin de pr\u00e9ciser l\u2019expression du g\u00e8ne d\u2019avirulence et des g\u00e8nes co-exprim\u00e9s\u00a0; (<em>iii<\/em>) une approche de g\u00e9nomique fonctionnelle pour tenter d\u2019exprimer la prot\u00e9ine correspondante dans un syst\u00e8me h\u00e9t\u00e9rologue et de rechercher son interactant dans les cellules v\u00e9g\u00e9tales. Monthly net salary about \u20ac1400.<\/p>\n<p><strong>Thesis summary<\/strong><\/p>\n<p>The objective of this thesis is to study the mechanisms underlying the evolution of pathogenicity of the poplar rust fungus, Melampsora larici-populina. Through a genome-wide association study (GWAS), we recently identified a locus in the genome of M. larici-populina, which could correspond to the avirulence locus Avr7, whose mutation is responsible for R7 resistance breakdown in poplar. The objective of the thesis will be to study in fine details this genomic region and to functionally validate this avirulence locus. This would be the first avirulence gene identified in this biotrophic fungal species. Several complementary approaches will be engaged: (i) a comparative genomics approach in order to list the polymorphisms in this region; (ii) a transcriptomic approach to clarify the expression of the avirulence gene and the co-expressed genes; (iii) a functional genomic approach to attempt to express the corresponding protein in a heterologous system and to search for its interactant in plant cells.
